Autophagy (cellular self-cleaning)

Autophagia

DE FR EN

The cell’s self-cleaning — biologically well established, but often over-interpreted in everyday claims.

Also known as: autophagy · cellular cleanup · autophagocytosis

Autophagy is the cell’s self-cleaning: it spots damaged proteins and worn components, wraps them, digests them in the lysosome and reuses the blocks. This quiet housekeeping runs constantly and steps up when energy is scarce — eating breaks, movement, sleep. In animal models good autophagy is linked to healthy ageing; Yoshinori Ohsumi won the 2016 Nobel Prize for the basics. That’s the solid side. In humans you can’t measure autophagy in daily life, and how much a given fasting window truly boosts it isn’t cleanly proven. Much sold online as an “autophagy boost” (certain supplements, extreme fasts) is overstated. The calm, dependable core: regular eating breaks instead of constant snacking, movement and good sleep are good for the body — whatever share ends up called “autophagy”. Note: fasting isn’t for everyone (see below). Tap a step in the graphic.

Levers & pathways

studiedGentle overnight break

Eat a bit earlier in the evening, later in the morning — a 12–14 h overnight window is simple and safe for most.

studiedDon’t snack constantly

Clear meals with real breaks between them — the calm, everyday core.

studiedBuild in movement

Walks, stairs, some endurance or strength — the most reliable nudge, good for everything else too.

studiedListen to your body

A little hunger is fine; exhaustion, weakness or fixating on food is not. The goal is wellbeing, not strictness.

infoNumbers and timeframes

In the laboratory, autophagy can be made visible in cells; in humans there is so far no simple blood value for it. The figures in circulation — that it starts after sixteen or twenty-four hours of fasting — come from animal experiments and do not transfer directly. What has been measured in humans: insulin falls in the first twelve to sixteen hours, ketones rise from about twelve to eighteen hours, and the growth switch mTOR is damped by a break from protein.

infoWhat the medicine does — and what it does not

There is no drug that switches on autophagy in humans in a targeted way. Rapamycin and metformin act on the switches and have been studied in ageing processes; no application for healthy people has been derived from that. Spermidine and resveratrol act in the laboratory on the same pathways; the human data are thin. What measurably pulls on the switches is the break from eating itself — insulin falls in the first twelve to sixteen hours, and the growth switch mTOR is damped.

Possible complaints

associatedFasting / eating breaks

The best-known trigger. Mechanistically clear, exact human magnitude still open. Gentle windows (e.g. 12–14 h overnight) are practical and low-risk.

studiedMovement & exercise

Physical activity demonstrably ramps up cell cleanup — perhaps the most solid lever, with plenty of side benefits.

associatedSleep

Sleep hosts many repair and cleanup programmes. Good sleep belongs here.

studiedWhat can realistically be expected

Autophagy is finely regulated — “more and longer” isn’t automatically better. It cures nothing on demand; it’s one building block of healthy habits.

studiedWho should be cautious

Fasting is NOT for everyone: underweight, eating disorder (or tendency), pregnancy/breastfeeding, children/teens, and diabetes on glucose-lowering meds — discuss with a doctor first.

studiedMitophagy

The special case: targeted recycling of tired mitochondria. Links cleanup with the energy level.

Blood values

studiedInsulin (fasting)

Falls during eating breaks — the state in which autophagy tends to rise. Not an autophagy test, just context.

associatedFasting glucose

Reflects the fasting/energy state. No direct autophagy measure exists in routine labs.

Nutrients

associatedSpermidine

A polyamine that can stimulate autophagy — linked in observational studies to healthy ageing. Human studies are only at the beginning.

associatedProtein timing

Lower-protein phases reduce growth signalling. Important: don’t stay too low on protein with age (muscle protection).

associatedMagnesium & B vitamins

Building blocks of a healthy energy metabolism in which cell cleanup works well.

Supplements

associatedSpermidine

The most-discussed supplement; early human trials ongoing. As an add-on, not a replacement — with realistic expectations.

traditionalResveratrol / berberine

Act in the lab on the switches (sirtuins/AMPK). In humans there are few studies on autophagy.

traditionalNMN / NR (NAD⁺ precursors)

Heavily marketed around “longevity”. In humans there are still few studies; in the lab the pathways are described.

infoMagnesium (glycinate/citrate)

Effective when deficient; effects on sleep and stress plausible but moderate. Too much causes loose stool.

Hormones

studiedInsulin ↓ & glucagon ↑

The fasting signature: low insulin, more glucagon — favours the cleanup.

associatedIGF-1 / growth signal

Less growth signalling shifts the cell from “building” toward “cleaning”.

associatedCortisol / stress

Brief stress (exercise, cold) can stimulate; chronic stress is rather unfavourable.

Food

associatedSpermidine-rich food

Wheat germ, aged cheese, natto/soybeans, mushrooms, legumes, wholegrains.

associatedCoffee

In animal models an autophagy inducer; in humans a plausible small contribution.

associatedGreen tea & olive oil

Polyphenols (EGCG, hydroxytyrosol) are linked to the process — part of good everyday food.

studiedLess constant snacking

Not what, but when: clear breaks between meals are the tangible part.

Alternative approaches — what people use

mixedIntermittent fasting

Cell cleaning starts in animal studies after about 12–16 hours without food. In humans the timing is not as clearly measurable as often presented — the figures from the mouse do not transfer directly.

studiedMovement & exercise

Endurance exertion triggers the same cellular processes as food deprivation — and is easier for most to sustain than long fasting phases.
